Holding onto power: why confidence intervals are not (usually) the best basis for sample size calculations

چکیده

Results With a little simplification, the sample size calculations for the traditional power-based approach, for equivalence studies, and following the new proposal can be shown to be much the same. The single fundamental difference is that the new proposal does not include a multiplier to increase the statistical power beyond 50% (i.e. only a 50:50 chance of detecting a true treatment effect of clinically important magnitude). The attempt to avoid having to define a minimum clinically important difference on a predefined primary outcome is wholly unsuccessful. The calculation of confidence interval width must be based on a particular outcome measure, still requires the size of an unimportant difference to be defined if the confidence interval is to exclude it, and additionally requires a likely true effect of treatment to be defined about which the confidence interval will be centred.

Outer and Inner Confidence Intervals Based on Extreme Order Statistics in a Proportional Hazard Model

Let Mi and Mi be the maximum and minimum of the ith sample from k independent sample with different sample sizes, respectively. Suppose that the survival distribution function of the ith sample is F ̄i = F ̄αi, where αi is known and positive constant. It is shown that how various exact non-parametric inferential proce- ′ dures can be developed on the basis of Mi’s and Mi ’s for distribution ...

Sample size calculations in clinical research should also be based on ethical principles

Sample size calculations based on too narrow a width, or with lower and upper confidence limits bounded by fixed cut-off points, not only increase power-based sample sizes to ethically unacceptable levels (thus making research practically unfeasible) but also greatly increase the costs and burdens of clinical trials. We propose an alternative method of combining the power of a statistical test ...

Sample size considerations for superiority trials in systemic lupus erythematosus (SLE).

For reasons of efficiency and ethics, sample size calculations are an important part of the design of all clinical trials. This paper highlights the statistical issues inherent to the estimation of sample size requirements in superiority trials particular to SLE. Calculations based on statistical power for testing hypotheses have historically been the method of choice for sample size determinat...

Area specific confidence intervals for a small area mean under the Fay-Herriot model

‎Small area estimates have received much attention from both private and public sectors due to the growing demand for effective planning of health services‎, ‎apportioning of government funds and policy and decision making‎. ‎Surveys are generally designed to give representative estimates at national or district level‎, ‎but estimates of variables of interest are oft...

Estimation and sample size calculations for matching performance of biometric authentication

Performance of biometric authentication devices can be measured in a variety of ways. The most common way is by calculating the false accept and false reject rates, usually referred to as FAR and FRR, respectively. In this paper we present two methodologies for creating confidence intervals for matching error rates. The approach that we take is based on a general parametric model. Because we ut...
